In vitro study | StemRegenin 1 treatment increased the number of CD34 cells with an EC50 of 120 nM after 5 to 7 days. Compared with the control group, mPB CD34 cells cultured with cytokines and SR1 (1 μm) for 7 days can increase the number of CD34 , CD133 and CD90 hematopoietic stem cells and progenitor cells by 2.6 times, respectively, 2.3 times and 10 times. Continuous culture with SR1 (1 μm) for 3 weeks resulted in an 11-fold increase in the total number of nucleated cells, a 73-fold increase in the number of CD34 cells, and a 1118-fold increase in the number of CD34 cells compared to controls. Treatment with SR1 (1 μm) cultured 1 × 10 SR1 (1 μm) could accelerate the proliferation of CD34 cells and reduce the expression level of VentX in human CD34 cells. Ectopic expression of VentX prevents sr1-induced CD34 cell enlargement. Continuous co-culture of bone morphogenetic protein 4 (20 ng/mL) with PGE2 (2 μm) and SR1 (0.75 μm) resulted in the formation of robust rhesus monkey induced pluripotent hematopoietic progenitors. CD34( )CD38(-)Thy1( )CD45RA(-)CD49f() cells were isolated based on CD34 expression and cultured in cells containing SR1 (0.75 μm) the number was expanded 3-fold in the culture medium and this long-term replanted hematopoietic stem cell phenotype was maintained. |
